Abstract (English)

The Introduction chapter reviews the literature on cyclic g-amino acids. The chapter covers their applications as small molecules and constituents of proteins.
The Main part is divided into three chapters. In the first chapter, the design, synthesis, and conformational analysis of linear peptides with cis-configured 2-(aminomethyl)-3-phenylcyclobutanecarboxylic acid (cis-g-CBAA) are studied. Special emphasis was put on both cis-g-CBAA enantiomers and their influence on the conformation and pharmacological activity of short neuropeptide Y analogs.
The second chapter concentrates on the design and synthesis of 4-aminopyrrolidine-3-carboxylic acids (cis-b-APCAs). Moreover, their application as unnatural constituents of the short peptidic GPCR ligands, particularly NPY Y4 and NTS1,2 receptors ligands, was evaluated.
In the third chapter, the visible light-mediated Aza-Paterno Büchi [2+2]-cycloaddition for the synthesis of enantiopure diazabicyclo[3.2.0]heptan-2-ones, isoxazolidines, and g-amino acid derivatives is presented.

Das Einführungskapitel gibt einen Überblick über die Literatur zu cyclischen g-Aminosäuren. Das Kapitel behandelt ihre Anwendungen als kleine Moleküle und Bestandteile von Proteinen.
Der Hauptteil ist in drei Kapitel unterteilt. Im ersten Kapitel werden das Design, die Synthese und die Konformationsanalyse von linearen Peptiden mit cis-konfigurierter 2-(Aminomethyl)-3-phenylcyclobutancarbonsäure (cis-g-CBAA) untersucht. Besonderes Augenmerk wurde auf die beiden cis-g-CBAA-Enantiomere und deren Einfluss auf die Konformation und pharmakologische Aktivität von kurzen Neuropeptid-Y-Analoga gelegt.
Das zweite Kapitel konzentriert sich auf das Design und die Synthese von 4-Aminopyrrolidin-3-Carbonsäuren (cis-b-APCAs). Darüber hinaus wurde ihre Anwendung als unnatürliche Bestandteile der kurzen peptidischen GPCR-Liganden, insbesondere der Liganden der NPY-Y4- und NTS1,2-Rezeptoren, untersucht.
Im dritten Kapitel wird die durch sichtbares Licht vermittelte Aza-Paterno Büchi [2+2]-Cycloaddition zur Synthese von enantiomerenreinen Diazabicyclo[3.2.0]heptan-2-onen, Isoxazolidinen und g-Aminosäurederivaten vorgestellt.
