Zusammenfassung (Deutsch)

Das Epstein-Barr-Virus ist weltweit verbreitet. Die Erstinfektion führt in der Regel zu einer lebenslangen, asymptomatischen Persistenz und die Verbreitung des Virus wird durch wiederkehrende Phasen von Reaktivierung mit Produktion infektiöser Nachkommen sichergestellt. Für die Induktion der Virusvermehrung spielt das virale Gen BZLF-1 eine entscheidende Rolle. Sein Genprodukt Zta ist als erstes virales Protein nach Induktion der Vermehrung nachweisbar. Die Expression von Zta reicht in latent infizierten B-Lymphozyten aus, die Kaskade der Gene der Virusreplikation in Gang zu setzen.
In der vorliegenden Arbeit wurde der Einfluss regulatorischer HI-Motive im Promotor von BZLF-1 auf die komplexe Regulation dieses Gens unter anderem mittels Transfektionsexperimenten untersucht. Je nach Zelltyp und den vorhandenen Transkriptionsfaktoren oder der Nachbarschaft zu anderen Promotorelementen in verschiedenen Promotor- / Reporterkonstrukten konnten die HI-Motive sowohl positiv- als auch negativ-regulatorisch wirksam sein.
Mittels neu hergestellter rekombinanter EB-Viren wurde gezeigt, dass im Kontext des Gesamtgenoms von EBV die HI-Motive jedoch für die Expression des Transaktivatorproteins Zta und für die Virusvermehrung von essentieller Bedeutung sind.
Die durchgeführten Experimente führten somit schließlich zu der Vorstellung, dass die HI-Motive durch ihren zelltypabhängigen Einfluss auf die Basalaktivität sowie auf die autokrine Stimulation einen wichtigen Beitrag zur Kontrolle der Transkription liefern, die als wichtigste Stufe der Regulation des BZLF-1 Gens angesehen wird.

The Epstein-Barr virus (EBV) is a human herpes virus infecting most of the worlds population. Primary infection normally leads to life-long persistence of the virus in the host and spread in the population is ensured by periodical reactivation. The immediate-early gene BZLF-1, which encodes the transcriptional activator protein Zta, plays a key role in the disruption of viral latency. Zta is the first viral protein detectable after the switch to virus replication and in B lymphocytes expression of Zta is sufficient to induce the cascade of the genes of the lytic replication.
With this work we have demonstrated the function and the role of the regulatory HI-sequence motifs in the promoter of BZLF-1 for the gene's complex transcriptional regulation. Performing transient transfection assays we showed that depending on the type of cell and the context of other regulatory sequence elements next the HI-motifs, these motifs had both activatory and inhibitory influence on transcription.
Using recombinant EB-viruses we found that in the context of the whole viral genome the HI-motifs played an essential role for the expression of the trans-activator protein Zta and consequently for the lytic replication of the virus.
In summary, due to their cell type dependent influence of basal activity and on autocrine stimulation, these motifs are supposed to be involved in the transcriptional control of BZLF-1, which is thought to be the gene's most important step in regulation.
