Das E1B-55kDa-Protein von Adenovirus Typ 5 ist ein multifunktionelles Polypeptid, dessen onkogenen Aktivitäten verschiedene Wirkmechanismen zugrunde liegen, die zumindest teilweise über die Modulation des Tumorsuppressorproteins p53 vermittelt werden. Das E1B-55kDa-Protein von Adenovirus Typ 5 ist ein multifunktionelles Polypeptid, dessen onkogenen Aktivitäten verschiedene Wirkmechanismen zugrunde liegen, die zumindest teilweise über die Modulation des Tumorsuppressorproteins p53 vermittelt werden. Im Rahmen dieser Arbeit wurde die Bedeutung des Exportsignals und anderer Domänen für die transformierenden Eigenschaften des E1B-55kDa-Proteins untersucht. Dazu wurden Punktmutationen innerhalb der NES-Region und in benachbarte Bereiche des E1B-55kDa-Proteins eingeführt. Die E1B-Mutanten wurden dann zusammen mit E1A in klassischen Transformationsversuchen getestet. Dabei konnte gezeigt werden, daß Mutationen im E1B-NES die Focus-Bildung gegenüber dem Wildtyp-Protein verstärken, während Veränderungen im benachbarten Bereich zum vollständigen Verlust der kooperativen Eigenschaften des Proteins führen. Datenbankanalysen ergaben, daß der dem NES folgende Abschnitt interessanterweise ein SUMO-Konjugationsmotiv enthält. Funktionelle Analysen zeigen, daß Ad5 E1B-55kDa tatsächlich durch SUMO-1 in kotransfizierten Zellen modifiziert wird. Aminosäureaustausche im SUMO-Konjugationsmotiv verhindern die kovalente Verknüpfung von SUMO-1. Dies resultiert in einer ineffizienten Repression p53-stimulierter Transkription und geht einher mit dem vollständigen Verlust der transformierenden Aktivitäten des großen E1B-Proteins. Funktionelle Analysen zeigen, daß die Konjugation von SUMO-1 den effizienten Import des E1B-Proteins in den Zellkern vermittelt. Außerdem bewirkt die Überexpression von SUMO-1 in transformierten Zellen eine Relokalisierung des E1B-Proteins in nukleäre, punktförmige Strukturen. Da diese Strukturen auch PML enthalten, könnte die SUMOylierung des E1B-Proteins ebenso die intranukleäre Assoziation mit PODs steuern. Die Ergebnisse dieser Arbeit lassen den Schluß zu, daß die Inhibition von p53 durch E1B-55kDa in den PODs stattfindet und durch SUMO-1-Konjugation reguliert wird.

The E1B-55K-protein from Adenovirus serotype 5 is a multifunctional polypeptide and provides function for complete oncogenic transformation mainly through modulation of the tumor suppressor protein p53. In this work the effect of the nuclear export signal and a further neighbouring domain on the transforming activity of the Ad protein has been investigated. E1B-point mutants have been tested in classical transformation assays with kidney cells from newborne rats. It has been shown that mutations within the NES increased focus-formation compared to the wildtype protein, whereas point mutations within the neighbouring region resulted in complete loss of the cooperative functions. Sequence analyses have shown that this region contains a SUMO-1-consensus motif and further experiments revealed that Ad5 E1B-55K, but not a point mutant, is covalently modified by SUMO-1 in vivo. It has been demonstrated for the first time that SUMOylation is absulutely required to enhance focus formation of E1B-55K in conjunction with E1A and to inhibit p53-transactivation activity. Immunofluorescence analyses from established cell lines indicate that this posttranslational modification is also required for efficient nuclear accumulation and intranuclear targeting of the Ad protein to dot-like structures containing SUMO-1, p53 and PML. These data suggest that SUMO-1 modification may play a role in intranuclear targeting of the Ad protein to nuclear multiprotein complexes called PODs. Additionally it has been concluded that intranuclear targeting of E1B-55K to PODs is a prerequisite for efficient transformation of primary rodent cells and that inhibition of p53 transactivation by E1B occurs in the context of PODs.
