Abstract (German)

Die Familie der hnRN-Proteine wurde traditionell mit der Verpackung naszierender Prä-mRNA in Verbindung gebracht. Untersuchungen der letzten Jahre haben jedoch ergeben, dass Mitglieder der hnRN-Proteinfamilie über vielfältige Funktionen verfügen. In diesem Zusammenhang wurde in früheren Arbeiten erstmals E1B-AP5 als zelluläres Zielprotein des adenoviralen E1B-55 kDa-Polypeptids identifiziert.
Transiente Promotor/Reporter-Analysen dieser Arbeit zeigen, dass E1B-AP5 über den aminoterminalen Teil, analog zu SAF-A, die basale Aktivität verschiedener Promotoren signifikant verringert. Aufgrund der funktionellen und strukturellen Redundanz muss man annehmen, dass die intrinsische Repressoraktivität von E1B-AP5 über die Inhibition von TFIIH vermittelt wird. Dagegen konnte die für SAF-A berichtete Inhibition hormonregulierter Transkription für E1B-AP5 nicht bestätigt werden. Interessanterweise wurde in diesem Zusammenhang das BRD7-Protein identifiziert, welches wahrscheinlich die E1B-AP5-induzierte Inhibition von TFIIH moduliert. Da E1B-AP5 zudem über BRD7 einen trimeren Komplex zu acetylierten Histonproteinen ausbildet, könnte die Chromatinstruktur aktivierter Gene die Funktionen von E1B-AP5 beeinflussen.
Erstmalig wurde der Nachweis des alternativ prozessierten E1B-AP5-Transkripts erbracht, das für die aminoterminal verkürzte Isoform E1B-AP5-A kodiert. Transiente Promotor/Reporter-Analysen zeigen, dass E1B-AP5-A nicht an den initialen Vorgängen der Transkriptionsregulation beteiligt ist. Vielmehr sprechen die Ergebnisse für eine Mitwirkung am nukleären Export von mRNA. In Übereinstimmung dazu stehen biochemische und Immunfluoreszenz-Studien die zeigen, dass E1B-AP5, nicht jedoch E1B-AP5-A in den Matrix- und Chromatin-Subkompartimente des Zellkerns lokalisiert ist. Zusätzlich lassen die Ergebnisse zur Transformationssuppression und zur Verringerung des Zellwachstums darauf schließen, dass E1B-AP5, nicht jedoch E1B-AP5-A an der Zellzykluskontrolle beteiligt sein könnte.

Translation of the abstract (English)

The hnRN-protein family was traditionally ascribed to the packaging of nascend pre-mRNA-transcripts. However, investigations of the last years revealed, that members of the hnRN-protein family share multifunctional properties. In this context earlier studies led to the identification of E1B-AP5 as a cellular target of the adenoviral E1B-55 kDa polypeptide.
As a result of this work, transient promoter/reporter-analysis show that E1B-AP5, like SAF-A, is able to repress the basic activity of several promoters via the aminoterminal part. Due to the functional and structural redundance, it has to be assumed that the intrinsic repression-activity of E1B-AP5 is coupled to the inhibiton of TFIIH. In contrast, the inhibition of hormone regulated transcription as revealed for SAF-A could not be confirmed for E1B-AP5. Interestingly these studies led to the identification of BRD7, a protein which might modulate the E1B-AP5 mediated inhibition of TFIIH. Furthermore the chromatin structure of actively transcribed genes might influence the functional properties of E1B-AP5 since it forms a trimeric complex with acetylated histones via the interaction with BRD7.
For the first time an alternatively processed E1B-AP5-mRNA was confirmed, wich encodes the aminoterminal truncated isoform E1B-AP5-A. Transient promoter/reptorter analysis show that E1B-AP5-A is not involved in the initial steps of transcriptional regulation. In contrast the results indicate that it participates most possibly in the nuclear export of mature mRNA. Consistent with this results, biochemical and imunofluorescence analysis show that E1B-AP5 but not E1B-AP5-A is localized in the nuclear matrix and chromatin subcompartements. Additionaly the results of the supression of transformation and the inhibition of cellular growth led to the conlusion, that E1B-AP5 but not E1B-AP5-A might participate in the control of the cell cycle.
