MADDAM (Metalloprotease and disintegrin dendritic cell antigen marker) ist ein molekularer Marker zur Unterscheidung von in vitro differenzierten Makrophagen und dendritischen Zellen. MADDAM wird in Monozyten exprimiert und während der in vitro Differenzierung zu Makrophagen herunterreguliert. Dagegen wird MADDAM im Verlauf der in vitro Differenzierung von Monozyten zu dendritischen Zellen konstitutiv exprimiert und durch die terminale Differenzierung nochmals nach oben reguliert. Weiterhin wird die Transkription des MADDAM Gens in Monozyten durch 1,25-Vitamin D3 deutlich verstärkt.
Zur Charakterisierung der regulatorischen Mechanismen, die die Expression des MADDAM Gens kontrollieren, wurde zunächst der Transkriptionsstartpunkt und der proximale Promotorbereich von MADDAM bestimmt. Dieser Bereich umfasst eine Region von ca. 150 Basenpaaren und enthält Bindestellen für Transkriptionsfaktoren wie SP-1, SP-3, NF-kB und VDR/RXR. Mittels Reporter- und EMSA-Analysen konnte die Funktionalität dieser Bindestellen bestätigt werden. Weiterhin wurden auch Bindestellen für Transkriptionsfaktoren im ersten Intron des MADDAM Gens gefunden, von denen ein �Vitamin D response Element� (VDRE) mitverantwortlich für die Regulation der MADDAM Transkription nach Stimulation mit 1,25-Vitamin D3 sein könnte. Zwei weitere solcher VDRE�s befinden sich im proximalen Promotor, die möglicherweise zusammen mit dem VDRE aus Intron1 die 1,25-Vitamin D3 vermittelte Regulation steuern.
Durch Deletions- und Mutationsexperimente wurden in Reporteranalysen die SP-und NF-kB Bindestellen als essentiell zur Aktivierung der MADDAM Transkription definiert, allerdings zeigen Transfektionen mit Reporterplasmiden in primären Makrophagen und dendritischen Zellen geringe Aktivitäts-unterschiede. Dies deutet darauf hin, dass weitere regulatorische Mechanismen für die zelltypspezifische Expression verantwortlich sind.
Weitere Untersuchungen zeigten, dass MADDAM eines der wenigen Gene ist, dessen Expression wesentlich durch epigenetische Mechanismen reguliert wird. Die Methylierung von CpG Resten im Promotorbereich spielt dabei wahrscheinlich eine untergeordnete Rolle, da die DNA von verschiedenen Spendern und der Zellinie THP-1 am MADDAM Promotor nicht methyliert vorlagen. Vielmehr sind Modifikationen der Chromatinstruktur, die durch Acetylierung von Histonresten verursacht werden von entscheidender Wichtigkeit. In THP-1 Zellen konnte die MADDAM Expression durch alleinige Inhibition von Histondeacetylasen dosisabhängig induziert werden. Weiterhin zeigte sich ein deutlicher Unterschied im Acetylierungsgrad der Histone am MADDAM Promotor von primären Makrophagen und dendritischen Zellen.
Zur Histonacetylierung notwendige Kofaktoren wie CBP, PCAF und TAFIIp250 werden in dendritischen Zellen sehr viel stärker exprimiert als in Makrophagen. Die bessere Verfügbarkeit dieser Acetyltransferasen ist möglicherweise der entscheidende Unterschied, durch den dendritische Zellen die Expression des MADDAM Gens steuern.

Transcriptional regulation of MADDAM / ADAM19
MADDAM (metalloprotease and disintegrin dendritic antigen marker) is a molecular marker for the in vitro differentiation of monocytes into dendritic cells. MADDAM is strongly expressed during dendritic cell differentiation, whereas the in vitro differentiation of monocytes into macrophages is associated with a downregulation of MADDAM mRNA levels. Furthermore, MADDAM is transcriptionally upregulated in primary monocytes after 4h stimulation with 1,25(OH)2VD3, the physiologically active metabolite of Vitamin D.
To investigate the underlying mechanisms, we defined the 5�UTR, the transcriptional start site and the proximal promoter of the MADDAM gene. The TATA-less promoter is located in a CpG island, contains an initiator sequence and shows putative binding sites for several transcription factors, including NF-kB, SP factors and the Vitamin D receptor. Reporter gene analysis revealed a minimal promoter of 150 bp that is active in HeLa and THP-1 cells. Transfection of primary cells with the 150 bp fragment resulted in a weak promoter activity in macrophages and a threefold higher activity in dendritic cells. EMSA showed binding of SP1, SP3, NF-kB and VDR to the proximal promoter and mutation of these binding sites led to a decreased promoter activity in luciferase assays.
Beside the initial definition of the MADDAM promoter, we found that MADDAM is one of very few genes, whose expression is basically regulated by epigenetic modifications. Methylation of CpG dinucleotides seems to be of minor importance, since DNA of different donors and the cell line THP-1 is not methylated at the MADDAM promoter. On the other hand, acetylation of histone tails at the MADDAM promoter has major effects in transcriptional initiation. Inhibition of histone-deacetylases in THP-1 cells resulted in a dose-dependent upregulation of MADDAM transcription. Furthermore, we found high acetylation levels of histone H3 at the MADDAM promoter in dendritic cells, not in macrophages. Cofactors like CBP, PCAF or TAFIIp250 that influence the acetylation state of histone tails were found to be differentially regulated in dendritic cells, versus macrophages.
