Abstract (English)
A hallmark in atherosclerosis is the generation of lipid loaded macrophage foam cells. In this thesis the effects of the different atherogenic lipoproteins, enzymatically modified LDL (E-LDL) and mildly oxidized LDL (Ox-LDL) on cellular cholesterol, sphingolipid and glycosphingolipid (GSL) metabolism, raft microdomain generation and cell surface receptor expression during in vitro foam cell formation and HDL3 dependent deloading of human monocyte-derived macrophages (MDM) were analyzed. Total cell analysis of lipid loaded and deloaded MDM, using lipidomics, genomics and proteomics based on mass spectrometry, Affymetrix microarray analysis, Taqman RT-PCR, flow cytometry, image microscopy and western blot analysis, was applied to unravel key mechanisms underlying the regulation of cellular lipid influx, raft formation, lipid efflux and storage as well as surface expression of receptors involved in lipid uptake and processing.
In the first part of this thesis apoE3/3 homozygous MDM were analyzed. It could be demonstrated that E-LDL loading predominantly increased cellular cholesterol content, while Ox-LDL loading preferentially increased cellular ceramide (Cer) content. This may be due to a stronger mRNA upregulation of acid and neutral sphingomyelinase (SMPD1, SMPD2), neutral sphingomyelinase activation associated factor (NSMAF) as well as upregulation of glucosylceramidase during Ox-LDL loading compared with E-LDL loading. Ox-LDL in comparison to E-LDL also led to a higher cell surface expression of Cer and the Cer derived GSLs lactosylceramide (LacCer, CDw17), dodecasaccharideceramide (CD65s), globotriaosylceramide (Gb3Cer, CD77) as well as GM1-ganglioside. ApoE with its high affinity for Cer in contrast to apoA-I was mainly bound and internalized by Cer enriched Ox-LDL loaded cells. Ox-LDL preferentially induced cholesterol/Cer rich-membrane microdomains while E-LDL loading induced cholesterol/sphingomyelin microdomains as confirmed in situ by confocal microscopy and by mass spectrometry of isolated detergent resistant membranes.
Microarray and flow cytometry analysis of the monocyte innate immunity receptor cluster revealed an induction of the complement system and of Fc-gamma-receptors with E-LDL but not with Ox-LDL loading. Further Ox-LDL induced mRNA and protein expression of ABCA1 to a higher extent than E-LDL. ApoE protein expression and expression analyzed by flow cytometry was higher during E-LDL loading while apoC-I expression was higher during Ox-LDL loading.
It is concluded that core-depleted E-LDL and surface oxidized Ox-LDL particles originating from the same LDL pool exert differential effects on cholesterol and sphingolipid metabolism as well as on cell surface receptor expression associated with the formation of different raft microdomains and thereby may trigger different macrophage effector functions during atherogenesis.
In the second part of the thesis apoE3/3 homozygous MDM were compared with MDM of the apoE4/4 genotype and ABCA1 deficient (Tangier) macrophages using the same incubation protocol with E-LDL and Ox-LDL loading and HDL3 deloading, to search for genotype specific differences in lipid and GSL metabolism. GSL surface expression during Ox-LDL loading was higher in apoE3/3 compared with apoE4/4 and ABCA1 deficient macrophages. Quantitative TaqMan-RT-PCR during Ox-LDL loading demonstrated no upregulation of acid SMase and a higher upregulation of acid ceramidase in apoE4/4 compared to apoE3/3 macrophages which could be the reason for the lower GSL surface expression of apoE4/4 macrophages. E-LDL loaded apoE3/3 macrophages demonstrated also a higher GSL surface expression compared with macrophages of a patient with hyperlipidemia and coronary heart disease. The apolipoproteins apoE, apoC-I and CETP which are involved in cholesterol and lipid metabolism were also analyzed in apoE3/3, apoE4/4 and ABCA1 deficient macrophages. mRNA expression as well as the protein amount of secreted apoE and apoC-I was higher in apoE3/3 than in apoE4/4 macrophages. Additionally surface and intracellular expression of these proteins was different between the two different genotypes and ABCA1 deficient macrophages.

Translation of the abstract (German)
Hauptmerkmal der Arteriosklerose ist die Bildung von Lipid beladenen Makrophagen so genannten Schaumzellen. In dieser Arbeit wurden die Einflüsse der unterschiedlichen atherogenen Lipoproteine, enzymatisch modifiziertes LDL (E-LDL) und mild oxidiertes LDL (Ox-LDL) auf den zellulären Cholesterol, Sphingolipid und Glycosphingolipid (GSL) Metabolismus, die Raft-Microdomänen Bildung sowie der Zelloberflächenexpression von Rezeptoren während der in vitro Schaumzellbildung und der HDL3 abhängigen Entladung von menschlichen Makrophagen untersucht. Eine Analyse von Lipid beladenen und entladenen Makrophagen mit Hilfe von Lipidomics, Genomics und Proteomics basierend auf Massenspektrometrie, Affymetrix Microarray Analyse, Taqman RT-PCR, Durchflußzytometrie, Mikroskopie und Western Blot wurde durchgeführt. Ziel war es, Schlüsselmechanismen der Regulation von zellulärer Lipid Aufnahme, Raft Bildung, Lipid Abgabe und Speicherung sowie Oberflächenexpression von Rezeptoren welche an der Lipid Aufnahme und Prozessierung beteiligt sind aufzuklären.
Im ersten Teil dieser Arbeit wurden apoE3/3 homozygote Makrophagen untersucht. Es konnte gezeigt werden, dass E-LDL Beladung hauptsächlich den zellulären Cholesterolgehalt erhöht während Ox-LDL vorwiegend den zellulären Ceramidgehalt erhöht. Dies könnte durch eine stärkere mRNA Expression der aciden und neutralen Sphingomyelinase (SMPD1, SMPD2), dem neutralen sphingomyelinase activation associated factor (NSMAF) sowie der Glucosylceramidase durch Ox-LDL verglichen mit E-LDL Beladung bedingt sein. Ox-LDL im Vergleich zu E-LDL führt auch zu einer höheren Zelloberflächenexpression von Ceramid und den aus Ceramid entstehenden GSL Lactosylceramid (LacCer, CDw17), Dodecasaccharidceramid (CD65s), Globotriaosylceramid (GB3Cer, CD77) sowie GM1-Gangliosid. Im Gegensatz zu apoA-I, wird apoE mit seiner hohen Affinität zu Ceramid hauptsächlich von Ceramid reichen Ox-LDL beladenen Zellen gebunden und aufgenommen. Ox-LDL induziert hauptsächlich Cholesterol/Ceramid reiche Membran-Mikrodomänen während E-LDL Beladung Cholesterol/Sphingomyelin reiche Mikrodomänen induziert.
Analyse mit Microarray und Durchflußzytometrie des �monocyte innate immunity� Rezeptor Clusters zeigte eine Induktion des Komplementsystems und der Fc-gamma Rezeptoren durch E-LDL aber nicht durch Ox-LDL Beladung. Desweiteren induziert Ox-LDL die mRNA und Protein Expression von ABCA1 stärker als E-LDL. ApoE Protein-Expression und Oberflächenexpression bestimmt durch Durchflußzytometrie war höher durch E-LDL Beladung während apo-CI Expression durch Ox-LDL erhöht war im Vergleich zu MCSF Differenzierung.
Es wird gefolgert, dass E-LDL und Ox-LDL Partikel, welche vom selben LDL-Pool stammen unterschiedliche Effekte auf den Cholesterol und Sphingolipid Metabolismus ausüben sowie auf die Oberflächenexpression von Rezeptoren verbunden mit der Bildung von unterschiedlichen Raft Mikrodomänen. Dadurch werden unterschiedliche Funktionen von Makrophagen während der Arteriogenese gesteuert.
Im zweiten Teil der Arbeit, wurden apoE3/3 homozygote Makrophagen mit Makrophagen vom apoE4/4 Genotyp und ABCA1 defizienten (Tangier) Makrophagen verglichen, um Genotyp spezifische Unterschiede im Lipid und GSL Metabolismus aufzudecken. Die GSL Oberflächenexpression während Ox-LDL Beladung war in apoE3/3 Makrophagen verglichen mit apoE4/4 und ABCA1 defizienten Makrophagen erhöht. TaqMan RT-PCR zeigte durch Ox-LDL Beladung keine Hochregulation der aciden SMase und eine stärkere Hochregulation der acid ceramidase in apoE4/4 verglichen mit apoE3/3 Makrophagen. Dies könnte ein Grund für die geringere GSL Oberflächenexpression von apoE4/4 Makrophagen sein. E-LDL beladene apoE3/3 Makrophagen zeigten ebenfalls eine höhere GSL Oberflächenexpression verglichen mit Makrophagen eines Patienten mit Hyperlipidämie und koronarer Herzkrankheit. Die Apolipoproteine apoE, apoC-I und CETP, welche in den Cholesterol und Lipid Metabolismus involviert sind, wurden auch in apoE3/3, apoE4/4 und ABCA1 defizienten Makrophagen untersucht. mRNA Expression sowie die Proteinmenge an sezerniertem ApoE und ApoC-I war in apoE3/3 gegenüber apoE4/4 Makrophagen erhöht. Zusätzlich gab es Unterschiede bei der Oberflächen und intrazellulären Expression dieser Proteine zwischen den unterschiedlichen Genotypen und ABCA1 defizienten Makrophagen.
