Abstract (German)
Konformationelle Gleichgewichte von Proteinen sind für die Erfüllung von zahlreichen, zellulären Aufgaben von entscheidender Bedeutung. Durch sie sind Proteine z. B. in der Lage schnelle enzymatische Reaktionen durchzuführen oder für Signaltrans-duktionswege nötige Protein-Protein-Interaktionen einzugehen. Daher ist es von großer Relevanz, Kenntnisse über die konformationelle Dynamik von Proteinen zu erlangen, um ihre Regulationszyklen beeinflussen und die Proteinaktivitäten modulieren zu können.
Guaninnukleotid-bindende (GNB) Proteine müssen ebenfalls in der Lage sein, verschiedene Konformationen anzunehmen, um ihre Funktion als molekularer Schalter zu erfüllen. Neben dem bestehenden Gleichgewicht zwischen einem inaktiven, GDP-gebundenen und einem aktiven, GTP-gebundenen Zustand, verfügen sie über weitere konformationelle Zustände im aktiven Zustand, wie für Ras bereits detektiert. Innerhalb dieser Arbeit wurden für das Ras-verwandte Protein Arf1 ebenfalls mehrere konformationelle Zustände in der aktiven Form detektiert.
Die Zuordnung der NMR-Resonanzen des Peptidrückgrats von [delta17]Arf1(wt)-Mg2+-GppNHp wurde in dieser Arbeit ermittelt. Dadurch sind bessere Aussagen über die gesamte Dynamik von einzelnen Aminosäuren des Arf1-Proteins möglich. Diese ermittelte Zuordnung wurde auf die Alanin-Mutante übertragen. Die Analyse der [1H, 15N] HSQC Spektren von [delta17]Arf1(wt) und der Alanin-Mutante zeigte die größten DeltaDeltacomb-Werte für die beiden Schalterregionen, sowie den C und N Terminus. Diese Regionen können daher als Bereiche mit der größten konformationellen Dynamik im Protein angenommen werden.
Durch 31P-NMR und [1H, 15N]-HSQC-Messungen konnte für das Arf1(wt)-Protein das hauptsächliche Vorliegen des Zustandes 2 (nach Ras Nomenklatur) detektiert werden. Ein geringer Anteil des Zustandes 1S von 10 12 % ist im Wildtyp-Arf1 jedoch auch enthalten. Dieser Zustand 1S wurde so benannt, da er bzgl. seines Verhaltens gegenüber Nukleotidaustauschfaktoren der Serin-Mutante ähnelt. Die Alanin-Mutante zeichnet sich durch ein bestehendes Gleichgewicht zwischen dem 1S und dem 1A Zustand aus. Die Gleichgewichtskonstante wurde als 0,4 ermittelt.
Bei der Hochdruck-NMR handelt es sich um eine aussagekräftige Methode für die Untersuchung von konformationellen Prozessen in Biomolekülen. Diese Methode besitzt das Potential zur Kompression, Denaturierung und Deaggregation von Proteinen und vermag bestehende Gleichgewichte in Proteinen zu verschieben und dadurch wenig populierte Zustände nachzuweisen. Durch 31P-NMR-Hochdruckreihen konnte der anfangs im Arf1(wt) hauptsächlich vorhandene Zustand 2 hin zum Zustand 1S verschoben werden.
Die durchgeführten [1H, 15N]-HSQC-Hochdruck-Messungen erlaubten zudem einen differenzierteren Blick auf das gesamte Protein und zeigten besonders druckempfindliche Bereiche. Dabei handelte es sich zum einen um die beiden Schalterregionen von Arf1. Zudem fielen die Aminosäure-Bereiche von AS 24- 31 und AS 126- 129 auf, die für die Nukleotidbindung verantwortlich sind.
Weitere angewandte Methoden verfügten ebenso über die Fähigkeit, die bestehenden Gleichgewichte im Protein zu verschieben und deutlicher zu offenbaren. Dazu zählen die Zugabe des chaotropen Agenz Guanidiniumhydrochlorid und die Zugabe von Metall-Zyklen-Molekülen. Paramagnetisches Cu2+-Zyklen diente hier zur schnellen Identifizierung der Resonanzen des Zustandes 1. Eine Bindung von Cu2+-Zyklen erfolgte nur an die (T48A)-Mutante von Arf1. Diese liegt somit hauptsächlich im Zustand 1 vor. Eine Cu2+-Zyklen-Bindung konnte dagegen für [delta17]Arf1(T48S) und das Wildtyp-Arf1 nicht angenommen werden.
Für das Wildtyp-Arf1-Protein konnte eine Bindung an das Effektorkonstrukt huGGA1-GAT detektiert werden. Diese Komplexbildung zeigte sich sowohl durch eine starke Linienverbreiterung in den 31P NMR-Spektren, als auch mittels analytischer Gelfitration. Eine Bindung des Nukleotidaustauschfaktors ARNO-Sec7 konnte dagegen nicht beobachtet werden. Dies weist wiederum auf das hauptsächliche Vorliegen des Effektor-bindenden Zustandes 2 im Wildtyp-Fall hin.
Diese Untersuchungen zeigen deutlich die Beeinflussung der bestehenden konformationellen Gleichgewichte in der aktiven Form von Arf1. Dies stellt die Regulation des GNB-Zyklus in Aussicht, wenn durch drug screening kleine Moleküle identifiziert werden könnten, die selektiv bestimmte Zustände in der aktiven Form zu stabilisieren vermögen. Dadurch könnten relevante Arf1-Funktionen in ihrer Aktivität selektiv beeinflusst werden.

Conformational equilibria of proteins are of essential importance to fulfill their many, cellular tasks. With them, proteins are able to undergo fast, enzymatic reactions or to make necessary protein-protein-interactions for signal-transduction. So it is of great relevance to gain knowledge of the conformational dynamic of proteins to influence their regulation-cycle and to modulate the protein-activity.
Guanine nucleotide binding proteins must be able to exist in several conformations, to fulfill their talks as molecular switch. Beside the equilibrium between the inactive, GDP-bound state and the active, GTP-bound state, there are different conformational states present in the active state, like already detected for Ras. In this PhD-thesis such states were also detected for Arf in the active form.
The backbone-assignment of the NMR-resonances of [delta17]Arf1(wt)-Mg2+-GppNHp was done here. With this, the whole dynamic of some amino acids of the Arf1-protein could be investigated. This assignment was transferred to the [1H, 15N]-HSQC-spectrum of the Arf1(T48A)-mutant. The analysis showed the most significant changes of Deltadeltacomb-values for the switch-regions as well as for the C- and N-terminus. Consequently these regions can be assumed as regions with the most conformational dynamic in the protein.
With 31P-NMR and [1H, 15N]-HSQC-measurements the mainly presence of state 2 (Ras nomenclature) can be assumed in the wildtyp-Arf1. A presence of the state 1S can be assumed to 10- 12% as well. This state was named after the similar behavior to the serine-mutant. For the alanine-mutant an equilibrium between the states 1A and 1S with an equilibrium-constant of 0.4 could be detected.
High-pressure NMR is a powerful method for the investigation of conformational processes in biomolecules. This method has the potential to compress, denaturize and disaggregate proteins and to shift existing equilibria at ambient pressure and make the detection of low populated states possible. With 31P-NMR-high pressure series the existing state 2 in Arf1(wt) could be shifted to state 1S.
The [1H, 15N]-HSQC-high pressure measurements allowed a more differentiated view at the whole protein and showed the most pressure-sensible regions. For Arf1 this was the case for the switch regions and the amino acid regions 24- 31 and 126- 129, which are responsible for the nucleotide binding.
Other methods used here were applied to shift equilibria and to detect other state. Here the addition of guanidine hydrochloride or metal-cyclen-molecules could be named. Paramagnetic copper-cyclen was helpful to detect the resonances of state 1. Here binding of copper-cyclen could only be detected for the alanine-mutant, but not for the serine-mutant or the wild type-protein. So this indicated also state 1 for the (T48A)-mutant.
For the wild type-protein binding of the construct huGGA1-GAT could be assumed. The line-broadening in 31P-NMR-spectra was a clear hint for the complex-building, as well as analytical gel filtration. The binding of the guanine nucleotide exchange factor ARNO-Sec7 could be detected for the wild type. This also indicated state 2.
These investigations showed clearly the presence of different states in the active form of the GNB-protein Arf1 and how they can be modulated. This regulation of the GNB-cycle can announce the stabilization of different active state with the help of small molecules after drug screening in the near future. With this the selective influence on the Arf1-funktions could be possible.
