Abstract (German)

Glukokortikoide (GK) werden in der Therapie der Akuten Lymphatischen Leukämie (ALL) sowie weiterer maligner lymphatischer Erkrankungen eingesetzt. Ein schlechtes initiales Ansprechen auf GK ist prognostisch für einen schlechten Verlauf.
Das nicht verstoffwechselbare Glukoseanalog 2-Desoxy-Glukose (2-DG) sensibilisiert GK-empfindliche ALL-Zellen gegen sehr niedrige Dosen GK und macht GK-resistente Zellen für GK empfindlich. Der genaue Mechanismus der Synergie zwischen GK und 2-DG ist jedoch noch unklar.
Vorarbeiten haben gezeigt, dass die Behandlung von ALL-Zellen mit Dexamethason (DEX) zu einer verringerten Expression des mitochondrialen Glutamat/H+-Symporters (SLC25A22) und des mitochondrialen Dicarboxylattransporters (SLC25A10) führt. Hier untersuche ich die mikrosomale Fraktion behandelter Zellen mit einem proteomischen Shotgun-Ansatz um den Mechanismus weiter aufzuklären.
Ich zeige, dass die Behandlung mit 2-DG alleine und in Kombination mit DEX zu einer erhöhten Expression von NMT-1, für welches eine Rolle in der Apoptoseinduktion be-schrieben ist, und einer verringerten Expression von HADH, einem Enzym der Beta-Oxidation, führt.
Weiterhin führt die Inhibition der Glykolyse durch 2-DG zu einer sehr stark erniedrigten Expression von VDAC1. Es zeigt sich kein synergistischer Effekt mit DEX.
Somit bleibt der synergistische Effekt zwischen 2-DG und Dexamethason weiter unklar.

Glucocorticoids are used in the therapy of acute lymphoid leukemia (ALL) as well as and other lymphoid malignancies. An initial insufficient response to glucocorticoids has been shown to be an important prognostic marker for a bad outcome.
2-Deoxy-glucose (2-DG) is a glucose analog which is not metabolized and sensitizes ALL-cells to non-toxic doses of glucocorticoids and resistant cells to glucocorticoids. However, the underlying molecular mechanism of glucocorticoid-induced cell death and the synergism with 2-DG remains poorly understood.
Prior work has shown a downregulation of the mitochondrial glutamate/H+-symporter (SLC25A22) and the mitochondrial dicarboxylate carrier (SLC25A10) in response to treatment with the glucocorticoid dexamethasone (DEX). Here I analyze the microsomal fraction of treated leukemic cells with a proteomic approach to further elucidate the mech-anism.
I show that treatment with 2-DG alone and in combination with DEX leads to increased expression of NMT-1, which has been implicated in the induction of apoptosis, and a reduction of HADH, an enzyme of beta-oxidation.
I also show that inhibition of glycolysis by 2-DG is associated with downregulation of VDAC1. No synergistic effect with DEX can be shown.
The mechanism of synergy between 2-DG and glucocorticoids remains unclear.
