Myrrhe wird seit Jahrtausenden zur Behandlung verschiedener Krankheiten verwendet. Durch moderne Studien konnte gezeigt werden, dass Myrrhe bei einer Vielzahl von Indikationen tatsächlich wirksam ist und Commiphora myrrha vor allem bei Colitis ulcerosa im Tiermodell und in einer klinischen Studie positive Effekte hatte. Trotz dieser vielversprechenden Ergebnisse, sind die vorhandenen Daten zum Wirkmechanismus und der Zusammensetzung des Myrrheharzes weiterhin unzureichend. In dieser Arbeit soll daher ein kleiner Beitrag zum Verständnis der Wirkung der Droge auf Zellebene zu geleistet werden und Teile der Wirkung eventuell sogar auf Einzelverbindungen zurückzuführen. Als Testmodell wurde hierfür das Adhäsionsmolekül ICAM-1 gewählt, welches bei der Pathogenese der Colitis ulcerosa eine Rolle spielt und in Zusammenhang mit Myrrhe bisher nicht untersucht wurde. Ziel ist es aber auch das bekannte Inhaltstoffspektrum der Myrrhe durch neue Substanzen zu erweitern.
Zu diesem Zweck wurde durch Mazeration und Perkolation ein ethanolischer Extrakt (96%, V/V) aus dem Gummiharz von C. myrrha hergestellt. Durch eine Flüssig-flüssig-Verteilung konnte dieser in eine n-Heptan- (HEP) und eine Methanol-Fraktion unterteilt werden. Alle drei Fraktionen wurden anschließend in einem in vitro Assay getestet, in dem für den Gesamtextrakt eine milde (bis 81,6 ± 4,2%, 50 µM) und für die HEP-Fraktion eine stärkere konzentrationsabhängige Senkung (bis 43,2 ± 3,6%, 50 µM) der ICAM-1 Expression nachgewiesen werden konnte. In mehreren Fraktionierungsschritten erfolgte mithilfe des ICAM-1 in vitro Assays anschießend eine bioaktivitätsgeleitete Isolierung aus der HEP-Fraktion, bei der verschiedene chromatogaphische Methoden zum Einsatz kamen. Unter anderem wurde eine Flash-Chromatographie an Kieselgel, eine Centrifugal Partition Chromatography und eine präparative HPLC an einer Biphenyl-Phase angewendet.
In der Folge wurden aus der HEP-Fraktion insgesamt 38 Reinsubstanzen isoliert und deren Strukturen über ein- und zweidimensionale NMR-Spektroskopie sowie HRESIMS aufgeklärt. Außerdem wurden optische und spektroskopische Daten erhoben und die Reinheit der Isolate mittels HPLC-DAD bestimmt. Die isolierten Verbindungen sind entweder den Sesquiterpenen (Germacrane, Eudesmane, Elemane, Cadinane, Guajane) oder den Triterpenen (Cyloartane, Dammarane, 4-Demethyllanosterolene) zuzuordnen. Unter ihnen befinden sich fünf Verbindungen, welche erstmals für die Art C. myrrha beschrieben werden (29, 31-34) sowie sieben Verbindungen, die im Rahmen dieser Arbeit zum ersten Mal in der Gattung Commiphora nachgewiesen werden (1, 9, 11, 14, 16, 17, 37). Außerdem sind elf bisher unbekannte Verbindungen hier erstmalig beschrieben (4, 5, 21-26, 35, 36, 38). Darüber hinaus konnten Literaturangaben zu NMR- und anderen spektroskopischen Daten bekannter Substanzen an vielen Stellen ergänzt oder korrigiert werden (2, 3, 11, 18, 19, 31).
Besonders hervorgehoben werden sollen die Verbindungen 25 und 26, bei denen es sich um 9,10-Seco-Eudesmane handelt und die damit über ein bis dahin unbekanntes Kohlenstoffseklett verfügen. Mit 24 wurde außerdem eine Substanz isoliert, die sich durch eine Endoperoxidstruktur auszeichnet, welche für Commiphora ebenfalls neuartig ist. Durch Vergleich mit bekannten Sesquiterpenen konnten darüber hinaus Theorien zur Biosynthese dieser neuartigen Verbindungen aufgestellt werden.
In einem nächsten Schritt wurde eine Auswahl an Substanzen auf ihren Effekt in Bezug auf ICAM-1 untersucht. Die höchste Aktivität konnte für Verbindung 11 (IC₅₀ 44,8 µM) und die Referenzsubstanz Furanoeudesma-1,3-dien (IC₅₀ 46,3 µM) festgestellt werden, während ein Großteil der Isolate keinen Effekt in dem gewählten Testmodell zeigten. Dies lässt vermuten, dass die Wirkung des Extraktes auf synergistische Effekte vieler Verbindungen zurückzuführen ist, die in ihrer Gesamtheit die Aktivität der Einzelkomponenten übertreffen. Weiterhin konnten durch einen Vergleich strukturell ähnlicher Verbindungen einige Theorien zu Struktur-Wirkungs-Beziehungen der Sesquiterpene aufgestellt werden. Bemerkenswert war außerdem, dass keine der aktiven Substanzen über eine exozyklische Methylenstruktur verfügt, welche bei vielen anderen Sesquiterpenlactonen eine Interaktion mit dem Transkriptionsfaktor NF-κB erst ermöglicht.
Der Mechanismus der Hemmung von ICAM-1 aber auch der Einfluss, den dieser Effekt auf das Krankheitsbild Colitis ulcerosa hat, bedarf weiterer Forschung, um zu verstehen, wie Patienten in Zukunft weiter von dieser alten Arzneipflanze profitieren können.

Myrrh has been used to treat various diseases for thousands of years. Modern studies have shown that myrrh is actually effective in many indications and that the gum resin of C. myrrha shows positive effects, especially against ulcerative colitis, in animal models and in a clinical study. Despite these promising results, the data available on the mechanism of action and the composition of the myrrh resin are still insufficient. This work should therefore try to make a small contribution to the understanding of the drug's effect in vitro and possibly even attribute parts of the activity to individual compounds. The adhesion molecule ICAM-1, which plays a role in the pathogenesis of ulcerative colitis and has not yet been investigated in regard to myrrh so far, was chosen as target. The aim is also to expand the spectrum of known constituents of myrrh resin with new substances.
For this purpose, an ethanolic extract (96%, V/V) was produced from the gum resin of C. myrrha by maceration and percolation. By means of a liquid-liquid partition, the extract could be separated into an n-heptane (HEP) and a methanol fraction. All three fractions were then tested in an in vitro assay, in which a mild concentration dependent reduction of the ICAM-1 expression (up to 81.6 ± 4.2%, 50 µM) could be observed for the extract, whereas the HEP fraction even had a better effect (up to 43.2 ± 3.6 %, 50 µM). Subsequently, a bioactivity guided isolation was carried out for the HEP fraction based on the ICAM-1 in vitro assay. Fractionation was performed by various chromatographic techniques including silica flash chromatography, centrifugal partition chromatography and preparative HPLC on a biphenyl phase.
As a result, a total of 38 substances were isolated from the HEP fraction and their structures elucidated using one- and two-dimensional NMR spectroscopy and HRESIMS. In addition, optical and spectroscopic data were gathered and the purity of the isolates was determined by HPLC-DAD analysis. The isolated compounds were identified as sesquiterpenes (Germacrane, Eudesmane, Elemane, Cadinane, Guaiane) or triterpenes (Cyloartane, Dammarane, 4-demethyllanosterolene). Among them are five compounds that could be described for the species C. myrrha for the first time (29, 31-34), as well as seven compounds that are first detected in the genus Commiphora (1, 9, 11 , 14, 16, 17, 37). In addition, eleven previously unknown compounds are described here for the first time (4, 5, 21-26, 35, 36, 38). Furthermore, literature on NMR and other spectroscopic data of known compounds could be supplemented or corrected in many cases (2, 3, 11, 18, 19, 31).
Particular emphasis should be given to compounds 25 and 26, which were identified as 9,10-seco-eudesmanes and thus possess a previously unknown carbon sceleton. Furthermore, 24 is a substance that features an endoperoxide structure, which is also novel for the genus Commiphora. By comparison with known sesquiterpenes, theories on the biosynthesis of the novel compounds could be established.
In a next step, a selection of compounds was examined for their effect on ICAM-1. The highest activity could be determined for compound 11 (IC₅₀ 44.8 µM) and the reference substance furanoeudesma-1,3-dien (IC₅₀ 46.3 µM), while the majority of the isolates did not show an effect in the opted test model. This suggests that the activity of the extract is due to synergistic effects of many compounds, which in total exceed the activity of the individual components. Furthermore, by comparing structurally similar compounds, some theories on structure-activity relationships of sesquiterpenes could be established. It is also noteworthy that none of the active substances has an exocyclic methylene structure which, in the case of many other sesquiterpene lactones, enables an interaction with the transcription factor NFκB.
The mechanism of action on ICAM-1, but also the influence this effect has in ulcerative colitis, requires further research in order to understand how patients can benefit from this ancient medicinal plant.
